殺死與固定
利用藥劑迅速把細(xì)胞殺死，以保持材料本來(lái)的狀態(tài)和結(jié)構(gòu)。
常用的固定有F.A.A固定液、卡諾固定液等。將固定液放在具塞小瓶中，投入材料，蓋好瓶蓋。用F.A.A固定液固定，時(shí)間至少24小時(shí)。F.A.A固定液既是良好的固定劑，也是保存劑，因此材料可長(zhǎng)期保存在固定液中備用。卡諾固定液穿透力強(qiáng)，固定較小材料一般1~6小時(shí)即可，zui多不超過(guò)24小時(shí)。因卡諾固定液不能做保存劑，所以固定后要用95%、85%酒精浸洗，每級(jí)約20min.，然后保存在70%的酒精中備用。
（三）脫水
脫水的目的是除去組織中的水分，便于透明、浸蠟、包埋等步驟的進(jìn)行，因?yàn)檫@些程序中所用的藥品都不能與水混合，所以必須脫水。常用的脫水劑為酒精。脫水過(guò)程應(yīng)緩慢進(jìn)行，不能操之過(guò)急或時(shí)間過(guò)長(zhǎng)，以免組織變硬變脆或發(fā)生收縮現(xiàn)象。脫水采用等級(jí)脫水（系列脫水），即從較低濃度酒精開(kāi)始，逐漸替換到高濃度酒精。一般為：50%→70%→80%→95%（此時(shí)可加少許番紅，使組織著色便于包埋時(shí)定位）→*（兩次），每級(jí)停留時(shí)間2~4小時(shí)或更長(zhǎng)（*中時(shí)間宜短）。
用95%酒精配制各濃度酒精的方法：設(shè)原有酒精濃度為A，所需配制酒精濃度為B，則取Bml原有酒精，加上A-Bml蒸餾水，即可獲得Aml的B%的究竟。例如：95%酒精配成70%的酒精，則取70ml的95%酒精，加上25ml蒸餾水，則配成了95ml的70%的酒精。
